RESUMO:
Cassava (Manihot esculenta Crantz) belongs to the family Euphorbiaceae Juss., and to the genus Manihot Mill.,
is a very widespread crop throughout Brazil, and plays an important role in generating employment and income.
In this sense, this study aims to characterize the genetic diversity of cassava ethnovarieties maintained and
managed by traditional farmers involved in the Uarini flour chain through SSR molecular markers. This research
is part of the project: “Interdisciplinarity to strengthen the Uarini flour production chain - Ethnobotany and genetic
diversity of Manihot esculenta Crantz”, approved in BID N. 003/2020 – PAINTER/FAPEAM, coordinated by
Dr. Juliana Rodrigues Larrosa Oler. The research will be developed in the geographical delimitation of the area
determined by the Geographic Identification (GI) Uarini, which comprises the municipalities of Uarini, Alvarães,
Tefé and Maraã, located in the state of Amazonas. In this study, each municipality will be considered as a
population. Two young and fully expanded leaves will be collected from 20 accessions distributed by the 5 most
frequent local cassava varieties in the farmers' crops in each municipality, making a total of 80 individuals in
the four populations. Molecular analyzes will be carried out at the Laboratory of Plant Genetics and Molecular
Biology of the University Campus of Alta Floresta, UNEMAT. Total genomic DNA will be extracted from
approximately 100 mg of leaf material following the CTAB method described by Doyle & Doyle (1987) with
modifications for the species. The amplification reactions will be performed with 10 microsatellite markers
labeled with 6-FAM fluorochrome (blue) and HEX (green). Polymerase Chain Reactions (PCR), quantification
and genotyping will be performed. The analysis of genetic diversity among local varieties will be carried out
in two stages: considering the total number of individuals belonging to a single population and, later, at the
population level, considering the four sampled municipalities.
