RESUMO:

Currently, teak (Tectona grandis) is considered one of the most important forest species in the world for timber production. The increasing demand for timber is associated with its high quality, beauty, and versatility in applications. However, some diseases have caused significant losses in the teak industry, including teak canker associated with L. theobromae, making it impossible to trade affected individuals. The aim of this study is to establish a rapid and simple protocol for the detection and diagnosis of L. theobromae in teak wood using qPCR. This research will be conducted in laboratory settings as part of an experimental investigation in the research department of Unemat. Simple and rapid DNA extraction protocols will be employed to directly identify L. theobromae from collected samples, with adaptations for the use of the DNA extraction kit Wizard® Genomic DNA Purification Kit (Promega). The overarching objective of this research is to develop a fast, simple, and accurate protocol for the detection and diagnosis of Lasiodiplodia theobromae in infected teak wood using qPCR. The hope is to establish a straightforward protocol for the quick identification of L. theobromae in teak wood, allowing the genetic improvement program to develop management strategies for the crop, thus reducing losses caused by the pathogen.
