RESUMO:

Tectona grandis L. f is a forest species of high economic value, used in the manufacture of luxury furniture and shipbuilding. In Mato Grosso, this exotic species has adapted well to the climate, presenting a high productive potential. However, the teak crop has been threatened by Ceratocystis wilt, a fungal disease that can lead to loss of production. The objective of this project is to develop a quick and simple protocol for the identification of Ceratocystis fimbriata in teak wood using RT-qPCR. The work will be carried out between August 2022 and December 2023, in the laboratories of the Graduate Program in Genetics and Plant Improvement, in partnership with the company TRC – Teak Resources LTDA. For DNA extraction, simple and fast protocols will be used, with adaptations for the use of the DNA extraction kit Wizard® Genomic DNA Purification Kit (Promega). Confirmation of the quantity and quality of the extracted RNA will be performed by electrophoresis in a 1% agarose gel and will be compared with standard ʎ RNA (lambda), with a Qubit® 2.0 Fluorimeter (Invitrogen). The Reverse Transcription (RT) procedure will be used to generate the DNA that will be used in real-time PCR, which will allow the identification of the pathogen present in these samples. 20 primers and the fluorescent compound SYBR® Green will be used. It is expected to develop a simple and effective protocol for the rapid identification of Ceratocystis wilt in Teak, thus contributing to the genetic improvement program of this species.
